The following pages assess the handiness in the visualization functions of Imaris 2.7 and Imaris 3.0. While of subordinate importance in light microscopy of cellular non-periodic structures it is of outstanding importance in molecular structure analysis and the analysis of periodic structure.


1. The Test Picture

Test image "eva.ims" and "eva-square-int.ims" in Imaris2 format generated by GenerateFile.cxx:

3. Gallery View -> correct in Imaris 2.7 and 3.0 (same definition of gallery)

With additional layers to generate a 3D-stack, saved as ims2:


as layer 0 is defined as bottom layer, squares should appear behind Eva
(if you can read Eva, the squares should be covered partly by the name, as they are on bottom of the name-box).

=> in 3D squares should appear in front of Eva (if you can read Eva, then the squares should cover a part of it, as they are on top of the name-box)

 

4. Volume rendering + Isosurface

-> correct in Imaris 2.7 and 3.0

(raytrace with transparency 0) in Imaris 2.7 :
 

squares (layer 0 ) are behind Eva 
-> layer 0 is not on top, but on bottom of stack! 
=> definition of Imaris 2.7 is incorrect!

Full 3D rendering in Imaris3 (right-handed coordinate system):
  -> orientation is correct

 
 

5. Section view

-> correct in Imaris 3.0, incorrect in Imaris 2.7

Summary

  1. Imaris 3.0 is correct in all views and definitions !
  2. The 3D-view in both Imaris 2.7 and 3.0 is consistently right-handed.
  3. Isosurface in Imaris 2.7 produces the same orientation like Volume rendering => right-handed
  4. The gallery in both Imaris 2.7 and 3.0 has the same view.
  5. The gallery definition in Imaris 2.7 is incorrect: Layer 0 is NOT on the top of the 3D-stack, but on bottom (like in Imaris 3.0)!
  6. The section view in Imaris 2.7 is incorrect (it matches with the incorrect definition of laye
